     John was born circa 1684 in Monomoit, Barnstable County, Massachusetts. John married Mary (?) on 11 July 1706 in Eastham, Barnstable County, Massachusetts.2 John and his family lived on the farm that had been his father's. He was involved in the civic affairs of the community as he held many differing offices over the years. John's wife, Mary, died before May 1762 in Chatham, Barnstable County, Massachusetts, leaving him a widower. John made his will on 27 May 1762 at Chatham, Barnstable County, Massachusetts.

In the Name of God Amen the Twenty Seventh day of March Anno Domini 1762 I John Nikerson of Chatham in the County of Barnstable within the Province of Massachusetts Bay in New England yeoman being very weak in Body but of Perfect Sound Mind and memory Thanks be given to God therefore calling unto mind my mortality Knowing that it is Appointed for all Men Once to Die do make and ordain this my Last Will and Testament that is to say Principally and first of all I Give and Recommend my Soul into the hands of God that Gave it and my Body I Recommend to the Earth to be Buried in Decent manner at the Discretion of my Executors nothing doubting but at the General Resurrection I shall Receive the same again by the mighty Power of God And as Touching such worldly Estate as it hath Pleased God to bless me withal in this Life I give and Devise & Dispose of the same in the following Manner and form –
Imprimis – I give and bequeath to my Grand Son Daniel Nikerson of my son Elisha the one half of all that my part and parcel of meadow in S. Chatham which I bought of Mr David McCoel Deceased on the conditions following that is to say if he come and settle in this County within One year after my Decease then it shall be to him the said Daniel and his heirs and assigns forever if not it shall be for and I do hereby Give the Same to my Daughter Mary and her heirs forever she paying to my said Grand Son Daniel afore Named the sum of Ten pounds y thirteen shilling & 4 pence lawfull Money out of her own estate
Item: I give and bequeath to my Grand Son John Nikerson the son of my Son Elisha of all the westerly part of that parcel of Land and meadows adjoining to John Buck’s Land that is to say from the following bounds and range viz: Beginning at the bans that leads into said Meadow and Still Continuing the Creek so all the Lands and meadows from this Described Range to said Buck’s Lands and I do hereby allow to my daughter Mary the sole use and Improvement of said Lands and meadows for the space of five years after my Death and then if the said John be living as an Inhabitant in this County the said Lands and meadows shall be to him and his heirs and assigns otherwise then It shall be equally divided betwixt my Two Daughters Namely Mary and Elizabeth they paying to said John the sum of Six Pounds Thirteen shilling and four pence Lawfull money Item – I give to my beloved Daughter Mary Nikerson Two thirds parts of all those household Goods and moveable Effects within Doors and without of Every sort and of any kind excepting what is herein already disposed of and after all my Just Debts and Charges of all sorts are paid out of them that is to say out all my moveables that are not herein particularly Given by her to be enjoyed Provided She my Said Daughter do pay to my granddaughter Patience Young the Sum of Si Pounds Lawfull Money or the value thereof in movables as they shall b appraised and at her marriage or at her arrival to the age of One and Twenty Years and this said Six pounds I Give to my Granddaughter for her Portion out of my Estate and in Lieu of what her mother would have had, I also further give to my Said Daughter Mary all that my part of a woodlot near to Barnabas Taylars and and all my Pews in the meeting house and the one half of all other my Lands & meadows that I have not herein or otherwise heretofore Disposed of to her and her heirs and assigns forever.
Item: I Give Every Sort and Kind as aforesaid after Debts and Charges of all Sorts are paid as aforesaid and I Also further give to my said Daughter Elisabeth and her heirs and assigns the One half part of all my Lands and meadows that I have not herein Particularly otherwise disposed of -
And I Do hereby Constitute and appoint my Daughter Mary and my Nephew William Nikerson of said Chatham to by my Executors of this my Last will and Testament And I Do hereby utterly Disallow Revoke and Disannull all other wills, Testaments Legacies y Bequests y Executions by me in anywise willed or Named Ratifying this and no other to be my Last Will and Testament forWitness whereof I do hereto set my hand and Seal the Day and year above written

John departed this life in Chatham. His will was probated on 7 September 1762.
